A SUPREX Protocol for Membrane Proteins
Abstract:
A new protocol is being developed to apply SUPREX (stability of unpurified proteins from rates of H/D exchange), a new H/D exchange and mass spectrometry–based technique that was developed to quantitatively study protein folding and protein-ligand binding interactions, to membrane peptides and proteins. A derivative of the transmembrane segement of the M13 viral protein is the model peptide studied in this work. The peptide and the peptide in a LPC (lysophosphatidylcholine) micelle environment are characterized, and methods that separate the peptide from the micelle to allow for mass spectrometric analysis are explored.
