Synthesis of Largazole Analogues
Abstract:

The marine natural product largazole, isolated from the cyanobacterium Symploca, possesses high differential growth inhibitory activity, targeting cancer cells over nontransformed cells. This interesting biological activity has been partially attributed to the ability of largazole to selectively inhibit class I histone deacetylases (HDACs) over other classes of the same enzyme. However, isoform selectivity within class I HDACs can be improved. Here we report the synthesis of several largazole analogues that will be used to further characterize structure-activity relationships and ultimately aid the design of a more selective histone deacetylase inhibitor.
