TOWARDS THE SYNTHESIS, ISOLATION, AND CHARACTERIZATION OF A NOVEL CYCLIC DEPSIPEPTIDE ANTIBIOTIC, WAP-8294A2
- Abstract:
In the last two decades, bacterial infections have reemerged as a serious public health threat. Due to the multidrug resistance of new “superbugs”, new antibiotics with novel mechanisms of action are urgently needed. WAP-8294A2 is a novel depsipeptide antibiotic that was isolated from Lysobacter staphylocidin (FERM BP-4900) in Japan and showed significant in vitro and in vivo antibiotic activity against methicillin-resistant Staphylococcus aureus. This research describes the efforts toward the total synthesis of a WAP-8294A2 derivative, WAP-1 via manual solid phase peptide synthesis and the isolation and purification of natural WAP-8294A2 from the producing strain, Lysobacter staphylocidin.
