Thermodynamic Analysis of Membrane Bound Receptor-Ligand Interactions
- Abstract:
There exists a need for high-throughput protein-ligand binding assays that are amenable to the analysis of membrane proteins in their native environment. SUPREX has the ability to detect ligand binding using picomoles of unpurified proteins in a high-throughput manner. In an attempt to develop SUPREX into a general assay for membrane protein-ligand binding analysis, a model system of a series of seven nanobodies that bind to epidermal growth factor receptor (EGFR) was investigated. Thermodynamic information was collected for all the nanobodies with no ligand, but due to signal suppression issues in the mass spectra of nanobody in the presence of the membrane-bound EGFR, the binding affinity of only one nanobody to EGFR was calculated. In addition, a new technique termed SPROX was also used to detect ligand binding.
