Siderophore-Ionophore and Siderophore-Antibiotic Interactions
Abstract:

Here we present an investigation of the host-guest complexation of iron siderophores with various ionophores using ESI-MS. In order to calibrate the system, equilibrium constants were calculated for host-guest systems of crown ethers and potassium chloride, including 18-crown-6, benzo-18-crown-6, and dicyclohexano-18-crown-6 ethers. Interactions between ferrioxamine B and 18-crown-6 were also characterized via ESI-MS. In a separate investigation, the redox interactions between ferrioxamine B and the antimalarial drug, artemisinin, were studied using CV in order to elucidate the mechanism by which artemisinin enters and kills the cell. The biological, chemical, and medical implications of these results are discussed.
