Thermodynamic Stability Measurements on Proteins in Complex Mixtures: Fundamental and Practical Applications
- Abstract:
SUPREX, a thermodynamic stability measurement technique, was used to measure the stabilities of proteins in solutions designed to mimic molecular crowding. These stabilities were compared to stabilities in dilute aqueous solution. There was no significant difference in stability for α-lactalbumin and further study is needed for cytochrome c. These results taken together with others previously reported suggest an interesting hypothesis, that molecular crowding may stabilize some proteins and not necessarily stabilize others. Another part of this work elucidates signal suppression issues that must be addressed before analyzing the thermodynamic stability of large numbers of proteins in cell lysates using SUPREX.
